So I have been wanting to make it out to Big Bend for several years and figure this will be as good a time as any. My plan is to work part of the day on Thursday the 17th and aim to leave Arlington around 2:00pm(or sooner) for a marathon drive. I am planning to stay at a motel in Alpine or Marfa and head to the park early on Friday the 18th. This would give us 3 days in the park and drive home on Monday the 21st. I know not everyone will be able to take off work the days but if you can take off on Friday or Monday try to make this trip.
I don't really have any plans other than wheeling, exploring and camping.
Park entrance is $5 per person a day and primitive camp sites are $8 a day for the whole site. The website says 8 people per camp site but I doubt it would be a big deal if we had 9 or 10.
Food and gas are going to be the main expenses.

I called and talked with a lady at the park. She said the best place to enter the park is on the west side near Presidio. She said it would be best to reserve a camp site and if we get there late we can set up camp and then check in with the station the next day. She also said there are some sites that are a 2-3 hour drive from the entrance. Once we get to the entrance the ranger station is 27 miles down a dirt road that takes about an hour. The back country sites have a fire ring and picnic table so that is a plus.
**8/2/12 Update**
I reserved the Guale campsite(pronounced Walley). The park website says it is 4WD/High clearance accessible. There are actually 2 sites at this location. I reserved them both so we will have plenty of space and no worries about people camping near us. The site is the most remote site at the park so I would recommend getting there a few hours before dark. The site is $8 each a night so its not a huge expense but beer donations are appreciated.

It sounds like this place is a little historic store.
If you are coming from the west there are a handful of stations in Presidio just before you enter the park.
This below link has some good info about the park. Page 7 has a decent map. It shows the 2 sites I reserved and didn't realize they were kind of far apart. Once we get closer to the time I may cancel the reservation on 1 site depending on the number of people that actually go. Page 8 on that link has the following note "A special-use permit is required for travel on all BBRSP roads
2WDHC and beyond." I am sure these can be obtained at the ranger station in the park.
